Siti Nurul Afiqah binti Johari10-5-95
Atopic dermatitis (AD) is a highly pruritic, recurring
inflammatory skin disease. It usually develops in early childhood and is frequently seen in children with a personal history of respiratory allergy and/or a family history of atopic disease.
Barrier function of skin
Skin barrier dysfunction
1. Ceramide deficiency• Ceramides are main source of essential FA, a
key role in epidermal barrier function.• Acts as a water modulator and permeability
barrier• Decrease in ceramide level is main
attributable factor in barrier disrupted dry skin of AD
2. Filaggrin loss-of-function mutations
FLG mutations
Corneocyte deformation
Disruption of organisation of
extracellular lipid
Reduction in NMFsIncrease in pH
Impaired antimicrobial
properties (S.aureus)
