Almost all proteins have a peak at 280 n, therefore, quantitative analysis of protein can be performed with a UV/Vis spectrometer. An alternative quantitative analysis method is colorimetry using reagent (such as the Lowry method , Biuret method, etc.)

Calibration curve Quantitative analysis First of all, spectra of each protein concentration are measured to produce a calibration curve. Next, unknown samples are measured and quantitative analysis is performed using the calibration curve.

Amid I peak contains the secondary structure information (α-Helix, β- Sheet etc.)
Using this peak, the IR-SSE program calculates SSE by PCR ( Principal Component Regression) or PLS (Partial Least Square) method.
